Just noticed that some of the screws are in a really awkward position and I'd assume that I'll have to take the wheels off to get them off

. . . unless anyone has any magic tip! ;)

and can you just unscrew the bolts, take the mudflaps off and then screw the bolts back in or is there a nut on the otherside of the wheel arch?

does not involve taking the wheels off
yes just unscrew and take of then put screws back in

If you can't get to the screws because of the tools you use, you can get some more space for easier access if you turn the steering full to left when removing left side and the opposite for the right hand side.
